Endpoints’ Manufacturing Day brought together industry innovators for an insightful conversation on cartridge-based drug delivery devices and large-volume subcutaneous (LVSC) delivery, featuring perspectives from SHL Medical and Contract Development and Manufacturing Organization (CDMO) Grand River Aseptic Manufacturing (GRAM). The session explored not only the technical nuances of developing cartridge-based systems, but also the power of partnership in bringing LVSC solutions to market. 

Delivery systems: Innovation with usability in mind 

SHL Medical, a pioneer in self-injection devices, highlighted the Maggie® 5.0 mL cartridge-based autoinjector during the session. Incorporating SHL Medical’s proprietary Needle Isolation Technology (NIT®), Maggie 5.0 is designed to enable LVSC delivery without compromising usability. By eliminating manual needle attachment, the device aims to provide patients with a more intuitive and safer injection experience.  

As Vince Masciopinto, Customer Solutions Director at SHL Medical, explained, Maggie was developed in anticipation of future market needs. Its introduction reflects how SHL Medical invests early in device innovation and supporting infrastructure, enabled by the company’s vertical integration and deep industry expertise, to close industry gaps in large-volume autoinjector technologies. This approach aligns with SHL Medical’s broader strategy: combining engineering foresight with flexible development pathways, patient safety, and robust technology to address the shifting demands of at-home administration and biologic therapies.

Built with the market-proven NIT, Maggie 5.0 accommodates a wider variety of formulation characteristics and volumes that are not possible with pre-filled syringe-based autoinjectors.

Fill and finish: Excellence meets collaboration 

On the manufacturing side, GRAM provides fill and finish services for sterile injectable biologics, operating 500,000 square feet of GMP space to support drug development and scalable manufacturing. Tyler BeVier, Director of Drug Product Manufacturing at GRAM, emphasized the complexity of cartridge filling, noting critical factors such as equipment compatibility, sterilization techniques, stopper designs, and process validation.  

GRAM’s recent investments in 5.0 mL cartridge capacity respond directly to the growing demand for LVSC delivery, where larger format containment introduces nuances and requires additional control, from maintaining container closure integrity to managing headspace and minimizing risk of variability across the fill-finish process.
